1 Scope
This standard specifies the sample preparation method and gas chromatography-mass spectrometry for the determination of diazepam and methaqualone residues in animal derived food.
This standard is applicable to the determination of diazepam and methaqualone residues in pig muscle tissue.
2 Normative references
The following referenced documents are indispensable for the application of this document. For dated references, only the edition cited applies. For undated references, the latest edition of the referenced document (including any amendments) applies.
GB/T 6682 Water for analytical laboratory use - Specification and test methods
3 Principle
The residual diazepam and methaqualone in the sample are extracted with acetonitrile, degreased with n-hexane, purified by C18 column, determined by gas chromatography-mass spectrometry, and quantified by external standard method.
4 Reagents and materials
Unless otherwise indicated, all the reagents used in the following are analytically pure reagents; and the Grade I water specified in GB/T 6682 are used.
4.1 Reference substances of diazepam and methaqualone: Content ≥ 99.5%.
4.2 Acetonitrile: Chromatographically pure.
4.3 n-Hexane: Chromatographically pure.
4.4 Methanol: Chromatographically pure.
4.5 Methylbenzene: Chromatographically pure.
4.6 C18 solid phase extraction column: 500 mg/3mL, or equivalent.
