本标准第一法规定了酶-重量法测定食品中总、可溶性和不溶性膳食纤维的条件和详细分析步骤。本标准第一法适用于谷类、蔬菜和水果及其制品中总的、可溶性和不溶性膳食纤维的测定,但不适用于含低分子量的抗性麦芽糊精、寡果糖、低聚半乳糖、多聚葡萄糖和抗性淀粉等食品的膳食纤维的测定。本标准第二法规定了酶重量法-液相色谱法测定食品中含低分子量抗性麦芽糊精的总膳食纤维的条件和详细分析步骤。本标准第二法适用于含有抗性麦芽糊精的糖果蜜饯(含巧克力及制品)、粮食及制品、糕点、饮料、乳制品、肉制品和保健食品等食品中总膳食纤维的测定。本标准的第二法为“含抗性麦芽糊精食品中总膳食纤维的测定 酶重量法液相色谱法”,修改采用了国际分析化学家协会方法AOAC2001.03(2004年第18版)《含有抗性麦芽糊精食物中的总膳食纤维的酶重量法和液相色谱法测定》(Totaldietaryfiberinfoodscontainingresistantmaltodextrinenzymaticgravimetricmethodandliquidchromatographydetermination),主要修改了酶解用缓冲液,降低了酶用量,并简化了用于液相色谱法测定的试样的处理步骤,可测定食品中含低分子质量抗性麦芽糊精的总膳食纤维。
