1 Scope
This standard specifies the examination method for Shiga toxin-producing Escherichia coli (STEC) in foods.
This standard is applicable to the examination of STEC in foods.
2 Equipment and materials
In addition to the conventional sterilization and culture equipment in microbiological laboratory, other equipment and materials required are as follows.
2.1 Thermostatic incubator: 36°C ± 1°C and 44°C ± 1°C.
2.2 Refrigerator: 2°C ~ 5°C and -18°C ~ -20°C.
2.3 Balance: with a sensitivity of 0.001 g.
2.4 Homogenizer.
2.5 Vortex oscillator.
2.6 Thermostatic water bath: 50°C ~ 55°C; thermostatic metal bath: 99°C ± 1°C.
2.7 Centrifuge: with maximum rotation speed of 16,000g.

2.12 Aseptic conical flask with stopper: 250 mL, 500 mL and 1,000 mL.
2.13 Aseptic Petri dish: with a diameter of 90 mm.
2.14 Aseptic homogenizing bag: with filter screen and aseptic homogenizing cup.
2.15 Flat-cap 8-tube strip or 96-well PCR microplates.
2.16 Magnetic bead separation and enrichment device.
2.17 Micropipettors and tips: 0.2 μL ~ 2 μL, 2 μL ~ 20 μL, 20 μL ~ 200 μL and 100 μL ~ 1,000 μL.
2.18 10 μL aseptic inoculating loop.
2.19 1.5 mL sterile EP tube, sterile microcentrifuge tube and sterile 15 mL centrifuge tube.
2.20 0.22 μm sterile filter;
2.21 Automatic microbial biochemical confirmation system.
2.22 Matrix-assisted laser desorption/ionization-time of flight mass spectrometer (MALDI-TOF MS).
