GB 5009.24-2026 National food safety standard - Determination of aflatoxin M in food
1 Scope
This standard specifies methods for the determination of aflatoxin M1 and aflatoxin M2 in foods (hereinafter referred to as AFT M1 and AFT M2).
The first method (liquid chromatography) and the second method (liquid chromatography-tandem mass spectrometry) in this standard are applicable to the determination of AFT M1 and AFT M2 in milk and dairy products and foods for special dietary uses.
The third method (enzyme-linked immunosorbent assay) in this standard is applicable to the screening determination of AFT M1 in milk and dairy products and foods for special dietary uses.
Method I Liquid chromatography
2 Principle
The test sample is extracted with methanol or methanol-water, cleaned up with an immunoaffinity column, detected by liquid chromatography with a fluorescence detector, and quantified by the external standard method.
3 Reagents and materials
Unless otherwise specified, all reagents used in this method are of analytical grade, and the water is grade 1 water as specified in GB/T 6682.
3.1 Reagents
3.1.1 Acetonitrile (CH3CN): chromatographic grade.
3.1.2 Acetonitrile (CH3CN).
3.1.3 Glycerol (C3H8O3).
3.1.4 Methanol (CH3OH).
3.1.5 Sodium chloride (NaCl).
3.1.6 Disodium hydrogen phosphate (Na2HPO4).
3.1.7 Potassium dihydrogen phosphate (KH2PO4).
3.1.8 Potassium chloride (KCl).
3.1.9 Hydrochloric acid (HCl).
3.1.10 Petroleum ether (CnH2n+2): boiling range 30 ℃ ~ 60 ℃.
3.1.11 Tween-20 (C58H114O26).
3.2 Reagent preparation
3.2.1 Phosphate buffer solution (hereinafter referred to as PBS): Weigh 8.0 g sodium chloride, 1.2 g disodium hydrogen phosphate, 0.2 g potassium dihydrogen phosphate, and 0.2 g potassium chloride; dissolve in 980 mL water, adjust the pH to 7.2 ~ 7.6 with hydrochloric acid, then add water to 1000 mL, and mix well.
3.2.2 Acetonitrile-water (15+85): Take 85 mL water and 15 mL acetonitrile, and mix well.
3.2.3 Methanol-water (50+50): Take 500 mL water and 500 mL methanol, and mix well.
3.2.4 10% glycerol methanol solution: Weigh 1.0 g glycerol, add methanol to dilute to 10 mL, and mix well.
3.2.5 Phosphate buffer solution containing 0.5% Tween-20 (PBS-T): Weigh 5.0 g Tween-20, add PBS to dilute to 1000 mL, and mix well.
3.3 Standards
3.3.1 AFT M1 (C17H12O7, CAS: 6795-23-9): Standard of ≥98% purity or its solution.
3.3.2 AFT M2 (C17H14O7, CAS: 6885-57-0): Standard of ≥98% purity or its solution.
3.4 Preparation of standard solutions
Warning: The entire analytical procedure shall be carried out in the designated area. This area shall be protected from light (direct sunlight) and shall be equipped with a relatively independent workbench and waste storage device. Throughout the entire test, operators shall take appropriate protective measures in accordance with the requirements for handling highly toxic substances.
3.4.1 Standard stock solution (10.0 mg/L): Weigh 10.0 mg each of AFT M1 and AFT M2 (accurate to 0.1 mg) into small beakers, add acetonitrile to dissolve, transfer to a 1000 mL volumetric flask, dilute to the mark with acetonitrile, mix well, and calibrate the concentration using a spectrophotometer (see Annex A), to obtain a standard stock solution with a concentration of 10.0 mg/L. Transfer the solution to a brown reagent bottle and store at -18℃ or below, protected from light; valid for 12 months.
