试样以盐酸，硝酸、氢氟酸、硫酸分解，过滤；残渣以碱熔融。加氟化钠、氯化铵、氢氧化铵及二乙基二硫代氨基甲酸钠(铜试剂)进行沉淀分离。用甲基异了基酮-硫氰酸铵萃取锌， 使其与大量铁、铜、铝、锰、镍、镉等素分离。在有机相中加入1 (2-吡啶偶氮)-2-萘酚(PAN)与锌生成红色络合物，在波长548nm处，测其吸光度，借此测定锌。
